机械牵拉对自发性高血压大鼠肥大心室肌细胞连接蛋白43表达的影响

摘　　要：目的:观察持续牵拉前后自发性高血压大鼠(SHR)左心室肌细胞连接蛋白43(Connexin43,Cx43)表达水平的变化及链霉素对该过程的影响。方法:将26只SHR随机分为:对照组、牵拉组和链霉素加牵拉组。采用Langendorff灌流体外心脏,经左房将一球囊导管插入左室,膨胀球囊使左室舒张末压升高1.064kPa,持续牵拉40min后剪下左心室壁,用RTPCR方法检测Cx43mRNA水平。结果:牵拉组Cx43mRNA水平明显高于对照组(P<0.01);与之相比,链霉素加牵拉组Cx43mRNA水平降低(P<0.05),而与对照组相比差异无统计学意义(P>0.05)。结论:机械牵拉可引起肥大心肌Cx43水平上调,后者可能参与了肥大心肌异常电活动的发生。链霉素可有效抑制该过程的发生,因而可能具有潜在抗心律失常作用。Objective:To investigate the change of Connexin 43(Cx43) expression level in SHR left ventricular myocyte before and after static stretch and the effect of streptomycin on this procedure. Method:Twenty-six SHRs were divided randomly into control group、stretch group and strep-stretch group. In a Langendorff perfusion preparation, the isolated heart was inflated by an intraventricular balloon through left atrial to increase 8 mmHg end-diastolic pressure. After 40 minutes of stable stretch, the left ventricles were cut down and were the Cx43 detected mRNA leves. Results:the level of Cx43 mRNA was significantly higher in stretch group than in control group and was reduced in the strep-stretch group compared with the stretch group, there was no different between the strep-stretch group and control group.Conclusion:Mechanic stretch caused the up-regulation of Cx43 level in hypertrophied cardiomyocyte, the latter was probably involved in the abnomal electric activity in hypertrophied heart . Streptomycin could inhibit this procedure effectively and probably had potential anti-arrhythimia effect.
