摘 要:目的探讨苯并[a]芘(BaP)对猪主动脉内皮细胞JunN端激酶(JNK)磷酸化的影响。方法培养猪主动脉血管内皮细胞,分别以不同浓度的BaP染毒(0、0.1、0.5、1、5、10μmol/L)24h,用Western-blot法检测JNK磷酸化的改变。结果无BaP作用时,磷酸化JNK1/2基本不表达;随BaP浓度的升高,JNK1/2被激活的程度升高,其中磷酸化JNK1水平升高比较明显,升高峰值为BaP浓度1μM左右,随之逐渐降低;而JNK2被激活的程度稍小,表达比较平稳。结论不同浓度的BaP作用下,内皮细胞JNK1/2分别不同程度地被激活,可能是导致HSP70基因表达改变和细胞损伤机制之一。Objective To study the effect of benzo(a)pyrene(BaP) on Jun N-terminal kinase(JNK) phosphorylation of porcine aortic endothelial cells. Method Porcine aortic endothelial cells were isolated and cultured in vitro. The cells were treated with BaP at 0, 0.1, 0.5, 1.0,5.0 and 10μmol/ L for 24 h,and the change of JNK phosphorylation was detected using Western-blot. Results The phosphorylations of JNK1/2 were both at low level without BaP stimulation, but the JNK1/2 was activated with the increasing BaP concentration, and the phosphorylation level was highest with BaP at 1μmol/ L, then decreased. However, the JNK2 was gradually activated with the increasing BaP concentration. Conclusion The JNK1/2 in endothelial cells was differently activated at different BaP concentrations, which may be one of the mechanisms of HSP 70 gene expression change and cell damage.
