摘 要:目的:通过观察针刺及光信息等不同授时因子导引对金黄地鼠视交叉上核一氧化氮合酶表达的影响。方法:实验于2002-08/2004-10在成都中医药大学针灸推拿学院和四川大学华西基础医学与法医学院组织胚胎学和神经生物学教研室完成。实验动物为健康雄性叙利亚金黄地鼠90只,体质量110~140g,随机分为电针组、光脉冲导引组、8-羟基-丙胺-四氢柰组、捆绑对照组、电针+光脉冲导引组、电针+8-羟基-丙胺-四氢柰组。每组15只。金黄地鼠经驯化后进入自由运行状态,在恒定黑暗第3天于近似昼夜时间9:00,15:00,21:00分别给予电针、光脉冲、8-羟基-丙胺-四氢柰、捆绑、电针+光脉冲导引、电针+8-羟基-丙胺-四氢柰刺激,每个时间点5只动物。各方法治疗2h后用免疫组织化学方法和图像分析技术,检测各组金黄地鼠视交叉上核中一氧化氮合酶的吸光度值,并采用q检验方法进行统计学分析。结果:90只金黄地鼠全部进入结果分析。各组各时间点视交叉上核一氧化氮合酶表达情况:在近似昼夜时间15:00光脉冲导引组比捆绑对照组视交叉上核内一氧化氮合酶的表达显著增加(116.600±8.678,109.600±8.735,q=54.00,P<0.05);近似昼夜时间21:00光脉冲导引组比捆绑对照组视交叉上核内一氧化氮合酶的表达显著增加(136.800±5.762,101.000±9.407,q=35.80,AIM: To observe the effect of electroacupuncture or photic stimuli zeitgeber on the expression of nitricoxide synthase in suprachiasmatic nucleus of golden hamsters.METHODS:The experiment was finished in the School of Acupuncture and Massage of Chengdu University of Traditional Chinese medicine,and Department of Histology and Embryology and Neurobiology,West China School of Basic and Forensic Medicine of Sichuan University,from August 2002 to October 2004.A total of 90 Syria golden hamsters,weighing 110- 140 g were divided randomly into electroacupuncture,photic stimuli,8 hydroxy 2 (di n propylamino)tetralin,electroacupuncture + photic stimuli,electroacupuncture + 8 hydroxy 2 (di n propylamino)tetralin,and seizing control groups(n=15).Golden hamsters moved freely following domestication.On the third day of constant darkness,hamstes were treated by electroacupuncture,8 hydroxy 2 (di n propylamino)tetralin,photic stimuli,electroacupuncture+ 8 hydroxy 2 (di n propylamino)tetralin,electroacupuncture + photic stimuli and seizing at 9:00,15:00,and 21:00,respectively,and 5 hamsters for each time point. At hour 2 following the treatment,the absorbance(A) values of nitricoxide synthase in hamster suprachiasmatic nucleus were observed by using immunohistochemistry and image analysis method.The q test was used to test intergroup differences.RESULTS:All 90 hamsters were involved in the result analysis.The expression of nitricoxide synthase in suprachiasmatic nucleus increased obviously in photic stimuli group as compared with seizing control group at 15:00(116.600± 8.678,109.600± 8.735,q=54.00,P< 0.05),as well as at 21:00(136.800± 5.762,101.000± 9.407,q=35.80,P< 0.05);However,the expression of nitricoxide synthase in suprachiasmatic nucleus increased obviously in electroacupuncture and electroacupuncture+ 8 hydroxy 2 (di n propylamino)tetralin groups as compared with seizing control group at 9:00(115.833± 9.641,110.667± 0.210,79.833± 17.600,q=36.00,30.83,PCONCLUSION:The expression of nitricoxide synthase in hamster su
