17β-雌二醇对肾上腺素诱导的心肌细胞肥大及原癌基因c-fos表达的影响

摘  要:目的利用体外培养的乳鼠心肌细胞,观察17β雌二醇(17βestradiol)对肾上腺素(phenylephrine)诱导的心肌细胞肥大及其原癌基因cfos蛋白表达的影响。方法以培养的乳鼠心肌细胞为模型并分组给药后,用计算机图像分析软件测量心肌细胞表面积,免疫细胞化学方法检测心肌细胞原癌基因cfos的蛋白表达。结果17β雌二醇明显抑制肾上腺素诱导的心肌细胞表面积增大,同时减弱肥大心肌细胞原癌基因cfos的蛋白表达。结论17β雌二醇可抑制肾上腺素诱导的心肌细胞肥大,其作用机制可能和抑制原癌基因cfos的蛋白表达有关。Objective To observe the effects of 17β estradiol on phenylephrine induced hypertrophy and expression of proto oncogene c fos in cultured neonatal rat cardiomyocytes.Methods Using cultured neonatal rat cardiomyocytes as a model and after treating with different agents, surface area of cardiomyocytes was measured by computer photograph analysis software; the proto oncogene c fos protein expression was assessed with immunocytochemistry method. Results 17β estradiol inhibited obviouslyphenylephrine induced increase of surface area of cardiomyocytes; 17β estradiol reduced the proto oncogene c fos protein expression of hypertrophic cardiomyocytes. Conclusion 17β estradiol can inhibit hypertrophy of cardiomyocytes induced by phenylephrine and its mechanisms might be associated with the suppression of c fos protein expression.
