绵羊蓝舌病发病过程中病毒抗原定位及免疫活性细胞动态观察

摘　　要：用云南绵羊益舌病病毒（ＢＴＶ）人工摘种绵羊３０只，接毒后１～１５天．每天剖检２只，同时剖检健康对照羊２只，用免疫荧光和细胞化学技术对靶器官和免疫器官进行连续观察．在接毒后６～８天．绵羊血片中发现了荧光阳性反应红细胞．接毒后２天，口角毛囊上皮，７天，颊、鼻粘膜上皮棘细胞层棘细胞胞浆观察到病毒抗原，且越接近糜烂溃疡灶含病毒抗原的棘细胞和游走进来蚕噬抗原的巨噬细胞越多．在免疫器官从接毒后５～６天起就可见嗜酸性粒细胞增生，巨噬细胞胞浆有病毒抗原．且在嗜酸性粒细胞周围观察到荧光强度不一、均质、多形态的抗原－抗体复合物附着．免疫器官中免疫活性细胞在接毒后６～９天受损较重，Ｔ细胞数减少．Ｂ细胞中成熟型和衰竭型浆细胞大量增加．有分泌抗体能力的未成熟型浆细胞不能形成增殖高峰，１２天后，Ｔ、Ｂ细胞增殖均形成高峰．本文就上述观察结果与动物发病、耐过等临床表现的相关性进行了讨论．Abstract Thirty sheep were inoculateel with bluetonge virus.1 to 15 days after inoculation,two sheep were dissected each day,two healthy sheep as control.The target organs and the immune organs were continucously observed with immunofluoresence and cytochemistry methods.6 to 8 days after inoculation,fluorescence positive red cells were seen in blood smear.Virus antigens were observed 2 day after inoculation in the epithelia of mouth corner and hair-follide epithelia,7 day after inoculation in heckel cells of cheek and nasalmucosa epithelia.The nearer to erosion and ulcer sites,the more heckel cells contained virus antigens and maerophages were.5 to 6 days after inoculation,acidophilic cells increased in immune organs and virus antigens were observed in macrophages.Different density.uniform and various forms of antigen-antibody cccomplexes were adhered to acidophilic cells.6 to 9 days after inoculation,immunologically competent cells were seriously damaged,T cells decreased, mature and seuile B cells obviously increased, but inmature plasmocytes which can secret antibodies didnt reach the peak.12 days after inoculation,T and B cells proliferated and reached the peak.The correlation between above mentioned results and clinical changes, pathogenesis were discussed in this paper.
