摘　　要：目的:建立以反相高效液相色谱法测定两性霉素B血药浓度的方法。方法:色谱柱为C18,流动相为0.005mol/L乙二胺四乙酸二钠-甲醇(3∶7),流速为1.0ml/min,检测波长为380nm。结果:两性霉素B检测浓度线性范围为0.125～4.0μg/ml(r=0.9998),高、中、低3种浓度的平均回收率为93.5%,日内和日间RSD均小于3.6%。结论:本方法简便、快速、专属性强、重现性好,适用于临床两性霉素B血药浓度的监测。OBJECTIVE:To determine the blood concentration of amphotercin B by RP-HPLC method.METHODS:The determination was performed with C 18 as the chromatographic column.0.005mol/L EDTA-2Na-methanol(3∶7)was taken as the mobile phase with the flow rate at1.0ml/min and the detection wavelength set at380nm.RESULTS:The linear conˉcentration range for amphotercin B was0.125～4.0μg/ml(r=0.9998).The average recovery rate for the3(high,middle and low)concentrations of amphotericin B was93.5%.Both the intra-day RSD and the inter-day RSD were less than3.6%.CONCLUSION:The method is simple,fast,specific and reproducible,which can be used for the clinical monitoring of the blood concentration of amphotericin B.
