摘 要:目的:通过动物实验研究了解肝毒清颗粒对肝损伤的保护作用。方法:选用雄性Wistar大鼠60只,随机分为6组,即空白对照组、模型组、肝毒清颗粒大、中、小剂量组,乙肝宁阳性对照组,用硫代乙酰胺复制急性肝损伤模型,检测血清的ALT、AST活性和血浆内毒素(ET)含量以及TNF-α、IL-6水平并取肝组织做病理检查。选用雄性Wister大鼠120只,分组同上,用10%的CCL4溶液复制慢性肝损伤模型,取血测定ALT、AST、TP、ALB、GLO,计算ALB/GLO比例并取肝组织做病理检查。结果:硫代乙酰胺急性肝损伤时,肝毒清颗粒能使血清ALT、AST活性、ET含量、TNF-α、IL-6水平明显降低,与模型组比较差异显著(P<0.05);能减轻肝细胞变性、坏死、出血,促进肝细胞再生。CCL4慢性肝损伤时,可降低ALT,AST,升高血清TP、ALB的含量,与模型组比较差异有显著性(P<0.05);减轻肝脂肪变性、减少纤维组织增生、促进肝细胞再生。结论:肝毒清颗粒对急慢性肝损伤具有保护作用。Objective: To observe the effect of Ganduqing(GDQ) on liver injury caused by hepatitis. Methods: ①Acute hepatic injury indaced by thioacetamide: 60 male Wistar rats were divided into 6 groups,namely normal control group,model group, Ganduqing large,middle,small dose groups and Yiganning positive contrast group. The changes of blood serum ALT,AST activity and plasma endotoxin TNF-α and IL-6 level were measured. And liver tissue was examined pathologically; ②Chronic liver injury induced by Carbon tetra-chloride: 120 male Wistar rats were divided into 6 groups as above. The changes of blood serum ALT、 AST、 TP、 ALB、 GLO were measured and the proportion of ALB/ GLO calculated, and liver organ examined pathologically. Results: ①Plasma ET level and blood serum ALT,AST levels and TNF-α、IL-6 levels in therapentic groups were significantly lower than in model group when acute liver injury (P<0.05); ②In Ganduqing therapy groups blood serum ALT and AST were significantly lower than in model group and TP and ALB content were significantly higher than in model group when chronic liver injury (P<0.05). Pathological examination found that Ganduqing can lighten liver cell denaturation, putrescence and hemorrhage, and speed liver cell regeneration in acute injury, and lighten liver fat degeneration and reduce proliferation of fibrous tissue in chronic injury. Conclusion: Ganduqing granule can reduce the hepatic injury and protect the liver when the liver was acute or chronic injured.
