白芍总甙对佐剂性关节炎大鼠滑膜细胞功能和脾细胞增殖反应的影响

摘　　要：本文研究了白芍总甙（ＴＧＰ）对大鼠佐剂性关节炎（ＡＡ）滑膜细胞功能以及脾淋已细胞增殖反应的影响及其作用机理。结果表明，ＴＧＰ５０ｍｇ·ｋｇ－１·ｄ－１×１０ｄｉｇ可使ＡＡ大鼠滑膜细胞过度分泌白介素１，肿癌坏死因子（ＴＮＦ）和前列腺素Ｅ２（ＰＧＥ２）的功能恢复正常，继而下调滑膜成纤维细胞的增殖；吲哚美辛２ｍｇ·ｋｇ－１·ｄ－１×１０ｄｉｇ抑制ＰＧＥ２产生，但增加ＡＡ大鼠滑膜细胞分泌白介素１与ＴＮＦ，从而促进滑膜成纤维细胞的增殖。ＴＧＰ５０ｍｇ·ｋｇ－１·ｄ－１×１０ｄｉｇ恢复ＡＡ大鼠脾细胞过低的伴刀豆球蛋白Ａ（ＣｏｎＡ）增殖反应与其下调巨噬细胞产生一氧化氮和ＰＧＥ２有关；体外实验亦证明，高浓度（０．４——６．４ｍｇ·Ｌ－１）ＴＧＰ负调节脾细胞ＣｏｎＡ增殖反应与其促进巨噬细胞产生一氧化氮和ＰＧＥ２有关。Effects of total glucosides of paeony proliferative response of splenocytes from adjuvant(TGP)on the function of synoviocytes and the arthritis(AA)rats and mechanisms of its actions werestudied.TheresultS of this study showed thatsynoviocytes from AA rats released a higher level ofIL一1,tumor necrosis factor(TNF)and prostaglandinE2(PGE2)than that from normal control group。Thesynoviocyte culture supernatants of AA rats were ableto enhance the proliferation of synovial fibroblasts(SF)from AA rats in vitro。 but those of normal con-trol group did not have this effect The abnormal func-tions(i.e,,IL一1,TNF and PGE2 production as well asSF proliferation) of synoviocytes from AA rats wererestored to normal levels when treated with TGP 50mg·kg-1·d-1 ig ×10 d. That the SF proliferationmediated by the synoviocyte culture supernatants ofAA rats was further promoted when treated withindometacin 2 mg · kg-1·d-1 ig ×1 0 d appeared toresult from the inhibition of PGE2 synthesis and theenhancement of IL一1 and TNF production in thesynoviocytes from these animals。Our data also showed that the recovery of dimi-nished Con A一induced proliferation of splenocytesfrom AA rats treated with TGP 50 mg· kg1·d-1ig×10 d,might be accompanied by a reduction in nitricoxide(NO)and PGE2 production in macrophages。The down一regulation of higher doses of TGP(0.4一6.4 mg·L-1) on Con A一induced mouse splenocyteproliferation in vitro was associated with an increasein NO and PGE,production in macrophages.Theabove results provide experimental evidence for TGPto treat rheumatoid arthritis.
