摘  要:N-(4'-羧苯基)-4-羟基-3,5-二叔丁基苯甲酰胺对十四烷酰佛波醋酸酯刺激大鼠多形核白细胞生成过氧化氢的影响孙士勇,韩锐(中国医学科学院药物研究所,北京100050)新维甲类化合物N(4'-羧苯基)-4-羟基-3,5-二叔丁基苯甲酰胺[N-(4...The effect of N-(4'-carboxy-phenyl)-4-hydroxy-3,5-di-t-butyl benzamide(R8605)on H2O2 formation in tumor promoter 12 O-tetradecanoylphorbol 13- acetate(TPA)-activated ratpolymorphonuclear leukocytes(PMN)was investigated.R8605 showed a significant concentratioir-dependentinhibiton of H2O2 formation in TPA-activated PMN.The concentration of R8605 whch decreased H2O2 for-mation by 50%(IC50)was 7 μmol·L-1,It also exhibitedthe same inhibitory effect in the presence of exogenoussuperoxide dismutase and IC50 was 5 μmol·L-1.Thedirect interaction between R8605 and H2O2 was not iden-tified,but the inhibition of O2 production by R8605 inTPA-activated PM N was observed.These results sug-gest that R8605 may inhibit H2O2 formation mainlythrough blocking O2 production.The inhibiton ofR8605 on active oxygen species released from TPA-acti-vated PMN may be associated with its anti-promotingmechanism.
