摘  要:目的:分析Cajal间质细胞(interstitialcellsofCajal,ICC)体外培养的要点,研究ICC在体外培养过程中的形态学变化;方法:体外分离、培养小鼠小肠ICC,采用ICC特异的c-kit抗体进行免疫荧光鉴定,应用光学显微镜和激光共聚焦显微镜观察其在体外生长30d的形态学改变过程。结果:体外分离、培养的ICC,贴壁后24h呈三角形或梭形,有少量突起;体外培养72h的ICC即出现明显的长突起,并且可与周围ICC相互形成突触连接,7d以上的ICC可以形成广泛的连接。体外培养的ICC可以在体外长时间培养,并且未发现明显的形态学变化。结论:体外培养3d后ICC形成类似于体内的网络连接,即可用于下一步实验,并可以稳定培养至少30d以上。ICC在体外的成功培养有望为深入研究ICC提供新的手段和借鉴。Objective:To study the key points of the culture of interstitial cells of Cajal(ICC) and its morphologic changes -in vitro-. Methods: The ICC was dissociated from the ileal segment of mice and cultured in M199 medium. c-kit immunofluorescence antibody was used for its identification under the confocal microscopy. The morphologic changes were observed under the light microscope and fluorescence microscope. Results::ICC exhibit a few axis-cylinders after 24 h cultured-in vitro-,and the longer axis-cylinders could observed to build synapse with each other after 72 h . More widespread connection could be formed in 7 days. The ICC could be cultured -in vitro- at least 30 days in our study without obviously morphologic changes. Conclusion: ICC could long term cultured -in vitro- and exhibit the same morphologic characters of network as it’s -in vivo-. The method of ICC culture established by us furnishes a new means for more research of ICC.
