肺靶向顺铂白蛋白微球含量的衍生化测定法

摘　　要：采用衍生化原理，使二乙基二硫代氨基甲醚钠（ＤＤＴＣ）与抗癌药顺铂（ＣＤＤＰ）反应，致紫外吸收度增加，并依据肺靶向顺铂白蛋白微球研制及其质量评价要求，建立了两种含量测定方法。一法回收率９９．８８％、ＲＳＤ＝０．６９％，用于背景干扰小的微球释放度测定；二法回收率９６．１５％、ＲＳＤ＝１．３４％，用于干扰大的微球包封率测定。方法简便、快速，效果满意。In this methiod the principle of derivatization was employed.Sodium di-ethyldithiocarbamate(DDTC)reacted with anticancer agent cis-dichlorodiammine platinum(CDDP)to form Pt(DDTC)_2,absorption of which increased.To study bovine serum albumin mi-crospheres loaded CDDP for pulmonary targeting and to evaluate its qualities,we developed towmethods for determining CDDP. The recovery of method 1 was 99.88%,RSD 0.69%, whichcan be used to determine the CDDP release from microsperes when the back-ground interferenceis low,The recovery of method 2 was 96.15%,RSD 1.34%,which can be used to determine therate in capsulation when the back-ground interference is high. These methods are simple andrapid.
