摘 要:目的研究IL-18抗柯萨奇B3病毒(Coxsackie B3 virus,CVB3)的作用.方法将心肌细胞分为4组.感染组:用CVB3感染心肌细胞;干预组:心肌细胞感染病毒后加入IL-18(4~30 ng/ml);IL-18组:正常心肌细胞加入IL-18(8 ng/ml);对照组:正常心肌细胞.观察培养心肌细胞病变(CPE)及心肌酶谱乳酸脱氢酶(LDH)和磷酸肌酸激酶(CK)的变化.并进行IL-18最大无毒剂量实验.结果感染组感染病毒后第2、5天心肌细胞病变程度较其余3组明显,LDH和CK浓度明显升高,与其余3组比较差异有极显著性意义(P<0.01).IL-18浓度≥40 ng/ml对正常心肌细胞有毒性作用;在4~20 ng/ml范围内对感染心肌细胞具有保护作用.结论 IL-18可以抑制CVB3对心肌细胞的损伤,对CVB3感染的心肌细胞有保护作用.Objective To study the effects of IL-18 against Coxsackie B3 viruses (CVB3). Methods The myocardial cells were divided into 4 groups: Infection group: the myocardial cells were infected only with CVB3; Treatment group: IL-18 (4-30 ng/ml) was given after the myocardial cells were infected with CVB3; IL-18 group: IL-18 (8 ng/ml) was added into the cultured myocardial cells; Control group: normal myocardial cells. The changes in the cultured cell pathogenic effect (CPE),LDH and CK were observed. The maximum nontoxic dosage test of IL-18 was performed.Results In infection group,the CPE of myocardial cells was aggravated and the levels of LDH and CK were significantly increased on the second and 5th day after virus inoculation as compared with other three groups (P<0.01).≥40 ng/ml IL-18 showed toxicity to normal myocardial cells. During a range of 4-20 ng/ml,IL-18 had protective effects on the infected myocardial cells.Conclusion IL-18 could inhibit the damage of CVB3 to the myocardial cells and had protective effects against the CVB-3-infected myocardial cells.
