成人胰岛细胞的分离纯化及初步的临床应用

摘　　要：目的分离及纯化成人胰岛,以用于同种胰岛细胞移植.方法利用胶原酶灌注消化以及Ficoll分离液分层纯化的方法获得成人胰岛细胞,并应用于2例临床糖尿病患者.结果经双硫腙染色鉴定,分离纯化后的胰岛细胞活性达到80%～90%;且经胰岛素释放试验表明,实验中收获的胰岛具有良好的胰岛素分泌功能.经临床试验,1例糖尿病患者在输入此胰岛后,已脱离胰岛素治疗;另1例患者已停止服用降糖药.结论用胶原酶灌注消化及Ficoll分离液分层纯化的方法获得成人胰岛细胞是可行的.Objective To isolate and purify human pancreatic islet cells for islets transplantation. Methods The pancreatic islets were isolated by collagenase digestion and then purified by Ficoll density gradients. The islets were transplanted into two diabetes cases. Results The islet purity identified by DTZ was 80 %～90 % and the insulin release experiments demonstrated that the insulin secreting function of the islets was well. The infusion of the islets led to the withdrawal of insulin or hypoglycemic agents in diabetes patients. Conclusion The method of pancreatic islet isolation and purification adopted in this research is available.
