摘  要:目的观察CD2相关蛋白(CD2AP)mRNA在大鼠阿霉素肾病模型中的表达,探讨其在蛋白尿发生发展过程中的作用。方法建立大鼠阿霉素肾病模型,应用光镜、电镜观察不同时期(第2、4、6、8周末)肾组织的病理改变,并采用半定量RTPCR的方法,检测大鼠肾皮质中CD2APmRNA的表达。结果阿霉素注射后:(1)大鼠24h尿蛋白量逐渐增加,第2、4、6、8周末分别为(144.5±16.4)、(250.0±45.5)、(440.1±62.1)和(626.5±94.2)mg/24h,均显著高于正常对照组(24.3±6.5)mg/24h(P<0.001);(2)大鼠肾皮质CD2APmRNA的表达在第4、6、8周末分别达到正常对照组的1.7倍、2.5倍和1.9倍。CD2APmRNA的表达与尿蛋白量呈正相关(r=0.58,P<0.01)。结论阿霉素肾病模型大鼠肾皮质CD2APmRNA的表达与大鼠蛋白尿的发生发展具有一定的相关性。Objective To investigate the expression of CD2-associated protein (CD2AP) mRNA in rats with adriamycin nephropathy, and to explore the possible effect on the development of proteinuria.Methods Thirty SD rats, weighted from 140 to 160g,were randomly divided into the nephropathy model groups and normal control group. The nephropathy model was established by a single intravenous injection of adriamycin 6mg/kg.While the normal control group received the same volume of 0.9% sodium chloride. At the end of the 2 nd, 4 th, 6 th or 8 th week after injection, the rats were sacrificed and the kidneys were acquired. The renal histology was observed under light microscope and electronic microscope. The semiquantitative RT-PCR was used for detecting the levels of CD2AP mRNA.Results (1)CD2AP mRNA levels in the 4th,6th and 8th week groups were 1.7, 2.5 and 1.9 times those in normal control group respectively.(2)The expression of CD2AP mRNA was positively correlated with the amount of proteinuria (r=0.56,P<0.01).Conclusion The increased expression of CD2AP mRNA is to some extent related to the development of proteinuria in adriamycin nephropathy rats.
