摘  要:目的:研究IL6诱导相关新基因LX3在人多组织中的表达状态,并进行LX3蛋白质超家族分析,预测该新基因的功能.方法:用AP-PCR法制备32P标记的LX3的全长基因探针,Northernblot分析新基因LX3在MultipleTissueRNABlots(MTRB)上的表达,结合生物信息学方法进行新基因LX3表达蛋白质的超家族分析.结果:新基因在人8种组织中有不同程度的表达,其中在睾丸组织、脾脏中表达最高,而在脑和胃组织中表达相对较低.超家族分析显示新基因LX3与外周结合蛋白超家族成员2GBP具有很高的同源性.结论:新基因LX3与IL-6诱导的淋巴细胞增殖高度相关.AIM: To study the expression status of novel gene LX3 induced by IL-6 in human multiple tissues and to analyze LX3 protein superfamily so as to forecast the function of the novel gene. METHODS: Full length LX3 cDNA probe tagged by 32P was synthesized by means of AP-PCR. The expression of novel gene LX3 on multiple tissue RNA blots was analyzed (MTRB) by the method of Northern Blot. To obtain more functional information, we carried out further analysis of LX3 protein superfamily. RESULTS: The novel gene LX3 was expressed, to different degrees, in 8 human tissues, with strong expressiion in the spleen and spermary but weak in the brain and stomach. Superfamily analysis confirmed that LX3 protein shared the high homology with the protein named 2GBP, a member of periplasmic binding protein. CONCLUSION: There is a high correlation between LX3 and lymphocyte proliferation induced by IL-6.
