摘 要:目的:观察吗啡成瘾大鼠脑区内阿片μ受体的定位分布,研究糖皮质激素(地塞米松)控制阿片戒断症状的分子机制.方法:剂量递增腹腔注射吗啡建立大鼠成瘾模型,给予地塞米松干预,纳洛酮促瘾后观察戒断症状,采用免疫组化实验观察成瘾后阿片μ受体的分布定位变化.结果:吗啡依赖大鼠经地塞米松干预后,由纳洛酮催促戒断症状评分明显低于未经地塞米松处理组;吗啡成瘾后的阿片μ受体在各脑区的表达下调,尤以蓝斑、伏隔核、下丘脑、腹侧被盖区、海马等脑区明显.结论:吗啡成瘾后阿片μ受体在奖赏中枢和海马的表达下降.地塞米松能抑制吗啡依赖大鼠戒断症状及脑区阿片μ受体的表达.AIM: To explore the molecular mechanism of glucocorticoid in the treatment of opioid withdrawal syndromes and the localization of μ-opioid receptor. METHODS: Rats received administration of morphine by intraperitioneal injection with dose adding progressively to develop the addiction model. Twenty-four rats were randomly assigned into three groups and received different kinds of treatment. Naloxone was administrated to precipitate withdrawal syndromes. Immunohistochemical staining was used to detect the localization of μ-opioid receptor. RESULTS: The withdrawal syndromes in glucocorticoid group were alleviated more distinctly than those in morphine group. The expression of μ-opioid receptor protein was lower in hippocampus, hypothalamus, locus coreuleus, ventral tegmental area (VTA) and nucleus accumbens (NAc) of morphine dependent rats. CONCLUSION: Glucocorticoid produces some suppressing effect on the withdrawal syndromes of morphine dependent rats. The μ-opioid receptor may regulate the activities of rewarding systems by presynaptic mechanism.
