摘 要:本文建立了用反相高效液相色谱法测定阿昔洛韦血浓度的方法。流动相为4%甲醇水溶液,流速为2.0ml/min,固定相为RP─C18(5μm),检测波长254nm;以液━固萃取为前处理,扑热息痛为内标,阿昔洛韦与内标的保留时间分别为7.10和12.27min。本法最低检测限为3on,RSD<5%。操作简便,适合于阿昔洛韦在体内药物动力学研究。n RP─HPLC method for determining serum level of acyclovir was established. Acyclovir extracted by Sep─Pak pretreat─ment cartridge could be detected at 254nm..An RP-C18 was used as the stationary phase and a solution composed of methanol:water(4:96V/V)as the mobile phase with a flow rate of 2.0ml/min.P-acetylaminophenol was used as an internal stan-dard.The retention time were 7.10and 12.27min respectively.The limit of detection was about 3 ng.The peak area ratio of the acyclovir and the interal standard against acyclovir concentration was linear with a correlation coefficient of 0.9998 over method is simple and precise and usable for pharmacokinetic studies in human beings.
