摘  要:目的探讨塔斯品碱盐酸盐(TA)对成纤维细胞作用及促进伤口愈合的机制。方法体外培养L929成纤维细胞,经不同质量浓度塔斯品碱盐酸盐(0.05,0.1,0.2,0.4和0.5 μg·mL-1)作用后,以及加入0.4 μg·mL-1塔斯品碱盐酸盐作用不同时间(24,48和72 h)后,采用ELISA测定其自分泌转化生长因子-β1(TGF-β1)及表皮生长因子(EGF)的变化规律。结果塔斯品碱盐酸盐可促进成纤维细胞自分泌TGF-β1和EGF,且随质量浓度的增大,作用时间的延长而增强。结论塔斯品碱盐酸盐可促进TGF-β1和EGF的自分泌,促进成纤维细胞及炎性细胞的趋化性,促进细胞外基质的沉积,促进创面愈合。OBJECTIVE: To investigate the mechanism of enhancing wound healing by taspine hydrochloride (TA). METHODS: Fibroblasts were cultured in vitro, and TA was applied with different concentrations (0.05,0.1,0.2,0.4 and 0.5 μg&middotmL-1) and time (24,48 and 72 h) to influence their autocrine. The levels of transforming growth factor-β1 (TGF-β1) and epidermal growth factor (EGF) were determined by enzyme-linked immunoadsorbent assay (ELIAS). RESULTS: TA increased autocrine of TGF-β1 and EGF by fibroblasts, the action was reinforced with the increasing TA concentration and the extending of time. CONCLUSION: The above results indicated that TA increased autocrining of TGF-β1 and EGF, and accelerated the chemotaxis of fibroblast and inflammation cell, the deposition of extracellular matrix and wound healing.
