胎鼠嗅球分离培养出神经干细胞的研究

摘  要:目的研究胎鼠嗅球中分离培养产生神经干细胞(NSCs)的情况。方法取孕16~18d的孕鼠,取胎鼠的嗅球,用含10%FCS的培养基培养传代分离出的细胞,免疫细胞化学方法鉴定培养和传代出的细胞,对染色阳性的细胞计数检测纯度。结果P75染色阳性证明分离培养的细胞为嗅鞘细胞(OECs);来源于嗅球的细胞培养出的神经球行Nestin染色阳性证明为NSCs;传代OECs纯度为95% ̄98%。结论胎鼠嗅球中可分离培养出NSCs,OECs可稳定传代,可以作为移植用的种子细胞。[Objective]To research the separation and culture of NSCs from fetal rat's olfactory bulb. Tissues of olfactory bulb were taken from fetal rats which were in pregnant within 16~18 d. The cells were cultured with medium including cell factor and 10% fetal bovine serum respectively. The cultured and transfer of cultured cells were inspected with immunocytochemistry test. The positive cells were counted. Stain of P75 on cultured cells proved they were OECs. Cultured nerve bulbs which cells were originated from olfactory bulb were proved to be NSCs by Nestin stain. The purity of OECs was 95%~98%. [Conclusion] NSCs could be separated from fetal rats' olfactory bulb. The cultured cells separated from olfactory bulb were NSCs. OECs might be transfer stably in vitro as well, which can be used as seed cells in transplantation.
