摘  要:目的:利用聚丙交酯-乙交酯共聚物强度大和甲壳胺无纺布可塑性好的特点,制作出一种力学性能良好及空隙均匀的新型聚合物支架。方法:实验于2004-05/12在第一军医大学珠江医院全军儿科中心实验室和中山大学高分子研究所进行。将2倍甲壳胺无纺布质量的聚丙交酯-乙交酯共聚物溶解于氯仿中,聚合物浓度为20g/L,将甲壳胺无纺布反复浸泡其中、烘干,直至均匀交联。去除氯仿细胞毒后,所制作的支架,测定其力学性能及孔隙率。结果:①新型聚合物支架抗压缩强度为1.4~2.0MPa,弯曲强度达16.3MPa,剪切强度为48MPa。②新型聚合物支架孔隙率达到82%~86%,孔径在100~300μm之间。结论:使用此法制作的组织工程支架,强度和可塑性较好,三维结构稳定,各项参数可控制,可以制作较大型体积的支架。AIM:To fabricate a novel copolymer scaffold made by Poly(dl- lactide- co- glycolide)(PLGA) embedded with chitosan nonwoven cloth with good mechanical prop erties and well- proportioned spaces based on the available characters of stron g intensity of PLGA and good compliance of the nonwoven cloth. METHODS:The experiment was completed in the Central Laboratory for Pediatrics ,Zhujiang Hospital,First Military Medical University,and Research Institute of H igh Polymer,College of Chemistry and Chemical Engineering,Sun Yat- sen Universi ty from May to December 2004.Double qualities of PLGA as chitosan nonwoven cloth were dissolved in chloroform to prepare the solution with the concentration of 20 g/L,and then the chitosan nonwoven cloth was soaked in the solution repeatedl y,followed by drying till steady intersection.After disposal of toxic cells in c hloroform,the mechanical properties and porosity of the prepared scaffold were d etected. RESULTS:① The anti- compression intensity of the made novel scaffolds range d from 1.4 MPa to 2.0 MPa,the flexion intensity reached 16.3 MPa,and shear inten sity was 48 MPa.② The novel scaffolds made by this method had the porosity of 8 2% to 86% and their porosity ranged from 100 to 300 μ m. CONCLUSION:The scaffolds made in this way possess the stable three- dimensio nal microstructure with satisfactory intensity and compliance as well as control lable parameters,so they can be made in a large- scale volume.
