摘  要:目的寻找应用蒿甲醚预防血吸虫后,引起免疫保护作用的抗原分子,为血吸虫病疫苗研究提供新的候选抗原。方法感染血吸虫7天后的家免口服蒿甲醚,收集血清,用蒿甲醚预防后的免疫免血清筛选日本血吸虫(Schistosoma japonicum,Sj)成虫 cDNA 文库,并对阳性克隆的插入基因片段进行 PCR 扩增及测序分析。结果经三轮筛选,获8个阳性克隆。测序分折所获的这8个序列,其中5个序列分别与日本血吸虫三磷酸甘油醛脱氢酶基因(Sj GAPDH)基因同源,其余3个序列为日本血吸虫28kD 谷胱甘肽-S-转移酶(Sj.GST)基因。结论 Sj.GAPDH 和 Sj.28kDGST 可能是蒿甲醚杀伤血吸虫后,引起对再感染起免疫保护作用的抗原分子。Objective to understand and identify the molecules related to the serum antibodies induced by artemether after it protect the host from Schistosomule infection and provide some useful candidate antigens for devel- oping Schistosomiasis vaccine.Methods artemether administrated orally to rabbit in 7-day after it is infected with schistosoma japonicum cercariae.When the protective immunity is induced,Sera from rabbit were collected.The schistosoma japonicum adult worm cDNA library was immunoscreened with the sera.The inserts of positive clones were specifically amplified by PCR and sequenced after they were cloned.Result 8 positive clones were obtained after three rounds of screening,and represent 2 genes.There are glyceraldehyde 3-phosphate dehydrogenase (Sj.GAPDH) and 28kD glutathione s-transferase(Sj.GST) respectively.Conclusion Sj.GST and Sj.GAPDH may be the molecules released by Sj.when artemether kill the worm in vivo,which play the import roles involved in induced protective im- munity.
