摘 要:用RT-PCR技术检测澳大利亚杏仁树李坏死坏斑病毒,对92个待测样品的病毒RNA进行扩增和检测,其中有79个呈阳性,表明检验样品的染病率较高.RT-PCR技术检测病毒的方法灵敏、准确,是检测苗木是否感染病毒的有效方法之一.Prunus necrotic ringspot virus(PNRSV)occurred commonly in Australia almond (Prunus dulcis (Miller) D.A.Webb) is very harmful to development of Australia almond industry.To find seedlings or saplings that infected by PNRSV as early as possible,a rapid and convenient testing technique is needed urgently.In this paper,reverse transcription polymerase chain reaction (RT-PCR) technique was used for this task after extracting Prune Dwarf virus RNA by RNeasy plant mini kit (Quiagen Inc.) with some modification.The results showed that there are 79 positive samples among 92 tested samples based on RT-PCR,and it is confirmed that this technique is a sensitive and ideal tool for detecting prunus necrotic ringspot virus.
