摘  要:目的:观察急性胆道感染大鼠肝细胞原胆汁分泌的变化。方法:制作急性胆道感染大鼠动物模型,取其肝细胞进行原代培养,利用反向高效液相色谱(RP-HPLC)技术,检测肝细胞培养上清液中胆汁酸和UCB含量。结果:急性胆道感染大鼠肝细胞培养上清液中均可检出一定量的非结合胆红素(UCB),同时三种主要结合胆汁酸(GCA,GCDCA,TCA)分泌减少,与对照组比较,有显著性差异(P<0.01)。结论:急性胆道感染大鼠肝细胞分泌会发生明显改变,原胆汁分泌胆汁酸减少,胆色素代谢发生改变,对胆色素结石的形成具有一定的影响。Objective To investigate the changes of the prime bile secretion of rat hepatocytes with acute cholangitis Methods Acute cholangitis rat models were established and the technique of primary culture of rat hepatocytes and Kupffer cells in vitro and the technique of RP-HPLC were employed.Some component of bile acid and UCB in the supernate of cultured hepatocytes were measured.Results A certain concentration of unconjugated bilirubin(UCB) could always detected in all the supernate of cultured hepatocytes of the model group but none in the control group,and the concentration of glycochenodeoxucholic acid(GCDCA),glycocholic acid(GCA) and taurocholic acid(TGA) in the supernate of cultured hepatocytes were significantly lower in the model group than in control(P<0.01). Conclusion The bile primely secretion of hepatocytes im model rats will be changed,therefore the bile acid secretion can be reduced and the metabolism of bile bilirubin can also be changed,these changes can affected the formation of bilirubin gallstones.
