摘  要:通过比较在不同灭菌条件下籼稻成熟胚的灭菌效果和诱导率,选出最佳灭菌组合:70 %酒精3min+10 %次氯酸钠10 min+0 .1%升汞30 min。在诱导过程中,不同基因型材料的最适基本培养基不同,桂华占的最适基本培养基为MS培养基,油占8号的最适基本培养基为N6 培养基。在诱导培养基中添加10 mg/ L 的椰子汁,有利于胚性细胞团诱导及绿苗分化,以桂华占为例,胚性细胞团诱导和分化率分别比对照提高10 .2 7%和11.76 %。By comparing the effects of sterilization and induction rate of Indica rice mature embryos in vitro culture with different treatments, the best treatment was selected, which was 70% alcohol for 3min+10% NaClO for 10min+0.1% HgCl 2 for 30min.To the induction, different genotypes needed different basic media,the suitable basic medium for Guihuazhan was MS, and that for Youzhan No.8 was N6. Adding 10ml/L coconut milk into the inductive medium was helpful to increase the induction rate of embryogenic callus and the differentiation of green seedling. As a result, the induction of embryogenic callus and the differentiation of green seedling of Guihuazhan were higher than CK by 10.27% and 11.76%, respectively.
