摘 要:为了从细胞水平探索循环的免疫物质在蛋白尿发生中的作用,应用(35) ̄S和3 ̄H-亮氨酸双同位素标记技术,观察15例激素敏感型肾病综合征(简称肾病)患儿的外周血单个核细胞(PBMC)培养上清液对大鼠肾小球上皮细胞(GEC)合成含硫化合物的影响,结果表明:肾病极期未经激素治疗组(9例)培养上清明显促进GEC掺入(35) ̄S(P<0.01),激素治疗组(6例)与正常对照组比较无显著差异。提示激素敏感型肾病患儿PBMC的产物可影响GEC的生物活性。AbstractTo identify the role of circulating immunefactor(s) on the mechanism of proteinuna,thesupernatant of peripheral blood mononuclear cell(PBMC) was obtained from 15 steroid-responsive pa-tients with idiopathic nephrotic syndrome,and its ef-fect on sulfated compounds synthesis by cultured ratglomerular epithelial cell(GEC)was studied by (35) ̄S,3 ̄H-Leucine dual-isotope lablelling technlque.Theresult showed that the supernatant of PBMC from 9pateints without steroid treatment increased(35) ̄S up-take by rat GEC(P<0.01),while there was no signifi-cant differencc in(35) ̄S uptake between the supernatantof PBMC from 6 patients treated with prednisone(1.5~2.0 mg / kg / d)and 6 normal controls.These re-sults indicate that the products of PBMC from pa-tients with steroid-responsive nephrotic syndromecan affect the biological fonction ofcultured rat GEC.
