摘  要:目的:探讨人参总皂甙(SPG)对大鼠局灶性脑缺血的保护作用及其机制。方法:用线栓法制备大鼠大脑中动脉闭塞(MCAO)模型,用放射性同位素法测定大鼠脑微血管及脑组织中一氧化氮合酶(NOS)活性用氯化三苯四唑(TTC)染色,用图像分析仪测定脑梗死体积,显微镜下观察脑组织的病理学变化。结果:中药保护4h(I4G)组脑微血管中NOS活性〔(12.9±11.7)nmol·mg-1·min-1〕明显低于假手术(S)组〔(42.1±21.0)nmol·mg-1·min-1,P<0.01〕及脑缺血4h(I4)组〔(103.8±34.9)nmol·mg-1·min-1,P<0.001〕,脑组织NOS活性〔纹状体(47.5±11.2)nmol·mg-1·min-1,海马(65.3±8.9)nmol·mg-1·min-1〕明显高于I4组〔纹状体(36.6±7.4)nmol·mg-1·min-1,海马(49.9±9.7)nmol·mg-1·min-1,P<0.05和P<0.01〕并伴随脑梗死体积减小(P<0.01)及脑组织病理变化减轻。结论:SPG对MCAO大鼠有明显的脑保护作用,其机制可能是通过减少脑微血管中NOS活性及使脑组织中的NOS活性维持在一定水平上而实现的。Objective: To investigate the protective effect of traditional Chinese medicine, saponins of panax ginseng(SPG,人参总皂甙), on the focal cerebral ischemia in rat brain and its mechanism. Methods: The rat model of middle cerebral artery occlusion (MCAO) was established by threadthrombus method. The nitric oxide synthase(NOS) activity in the cerebral microvessels and tissues was determined by radioisotopic method. Triphenyltetrazolium chloride staining and image analysis equipments were used for the measurement of infarct volume.Pathological changes were observed by microscope with hematoxylin and eosin(HE) staining. Results: The NOS activity in the cerebral micro vessels of SPG ischemia 4 hours (I4G) group〔(12.9±11.7)nmol·mg^-1·min^-1〕 was lower than that of the shamoperation group 〔(42.1±21.0)nmol·mg^-1·min^-1, P<0.01〕 and ischemia 4 hours (I4) group 〔(103.8±34.9)nmol·mg^-1·min^-1, P<0.001〕. In I4G group, the NOS activity〔(47.5±11.2)nmol·mg^-1·min^-1 in striatum and (65.3±8.9)nmol·mg^-1·min^-1 in hippocampus〕 was higher than that of I4 group 〔(36.6±7.4)nmol·mg^-1·min^-1 in striatum and (49.9±9.7)nmol·mg^-1·min^-1 in hippocampus, P<0.05 and P<0.01〕, accompanied with the decrease of infarct volume(P<0.01) and less ischemic injury of pathology. Conclusion: SPG has remarkable protective effect on the brain of MCAO rat model. The mechanism could be by meaning of inhibiting the increase of NOS activity in the cerebral microvessels, meantime maintaining NOS activity in cerebral tissues at certain level.
