摘  要:目的:探讨实验性糖尿病家兔模型早期肺组织超微结构的变化及中药干预治疗对糖尿病肺损伤的影响。方法:40只雄性大耳白兔随机分为糖尿病组、中药治疗组和正常对照组。用四氧嘧啶(140mg/kg)空腹耳缘静脉一次性注入制备糖尿病家兔模型;成模后给中药治疗组家兔灌胃复方中药液6周;6周后杀检以透射电镜观察肺组织超微结构。结果:糖尿病组家兔肺泡上皮细胞及肺微血管内皮细胞间基底板较正常对照组增厚,蛋白沉积不均一,内皮细胞胞饮泡数量显著增多;型肺泡上皮细胞嗜锇板层小体囊扩大,大部分不含嗜锇板层小体。与糖尿病组相比,中药治疗组家兔肺微血管内皮细胞基底板无明显增厚,少量蛋白质沉着,内皮细胞胞饮泡较少;型肺泡上皮细胞粗面内质网池仍有囊性扩张,嗜锇板层小体嗜锇物质稍见增多。结论:肺脏也是糖尿病攻击的靶器官。早期采用中药干预治疗可减轻靶器官的损害。Objective: To investigate the changes of ultrastructure in lung tissues of early diabetic rabbits and therapeutic effects of traditional Chinese herbal medicine on lung injury induced by diabetes mellitus. Methods: Forty male white rabbits were randomly divided into diabetic group(n=15), herbstreated group(n=15) and normal control group.Diabetic model of rabbits was induced by a single intravenous injection of alloxan (140 mg/kg).After establishment of diabetic model, a decoction made by a compound prescription was poured into stomach for 6 weeks in herbstreated group. At the end of the sixth week, all of the rabbits were killed, and their lung tissues were observed under transmission electron microscope. Results: Compared with the control group, the following items were more obvious in the diabetic group than that in the control group, such as basal lamina of alveolar epithelial cells and pulmonary endothelial cells in diabetic rats were thicker, protein deposits were more various in density, the number of pinocytotic vesicles in pulmonary endothelial cells was apparently greater; the rough endoplasmic reticulum in typeⅡ alveolar cells was more enlarged and most typeⅡ alveolar cells did not contain osmiophilic lamellar body. However, in the comparison between the diabetic group and herbs treated group, in the treated group the basal lamina of pulmonary endothelial cells showed no significant thickness, few protein deposited, and number of pinocytotic vesicles in pulmonary endothelial cells was less; the moderate cystic dilatation of the cistern of rough endoplasmic reticulum in typeⅡ alveolar cells remained and osmiophilic materials in these bodies were increased in certain extent. Conclusion: Lung is the target organ of diabetic damage. Early herbal interventional treatment can alleviate the damage of target organ.
