摘　　要：目的建立植入用缓释MTX体外释放方法。方法选择体外释放的重要影响因素,采用四因素三水平的正交试验,以3个综合考核指标进行优选。结果释放介质为偏碱性等渗液;介质浓度为25%;介质体积为200ml;搅拌转速为50r·min-1。结论所定方法可用于植入用缓释MTX的体外释放测试。Aim To establish the release method of methotrexate sustained release for implant in vitro.Methods An orthogonal form of L_9(3~4) was adopted to observe the effects of four factors and three levels. The optimization of four experimental parameters was made according to the orthogonal results.Results The selection: The release menstruum is alkaline iso-osmotic solution; concentration of the release menstruum is 25%; cubage of the release menstruum is 200 ml; the rotate speed is 50 r·min~ -1 .Conclusion The method established is suitable to evaluate the in vitro release rate of methotrexate sustained release for implant.
