摘　　要：目的建立血浆中多塞平(doxepin,DXP)、氯氮平(clozapine,CZP)的HPLC检测方法。方法采用日本分光JASCO1500系列高效液相色谱仪,KromasilC18柱(150mm×4.6mm,5μm),以地西泮(diazepam,DZP)为内标,流动相组成为甲醇-乙睛-水-四甲基乙二胺-冰醋酸(50∶50∶100∶2∶1.5),流速为0.8ml·min-1,紫外检测波长254nm,灵敏度AuFs=0.02。结果血浆中两组分和内标均能很好地分离,无明显的干扰峰。线性良好,日内、日间RSD<8%。结论本法操作简便、结果可靠,适用于药物监测和中毒分析。Aim To develop a method for determining doxepin(DXP) and clozapine(CZP) in serum by high performance liuquid chromatography(HPLC)for providing a scientific basis for clinical use of hypnotic drugs. Methods 0.25 ml serum sample was extracted by Ether,dissolved by mobile phase. Kromasil C_ 18 (4.6 mm×150 mm,5μm)column was used;mobile phase: methanol-acetonitrile -water-C_ 6 H_ 16 N_ 2 -acetic acid (50∶50∶100∶2∶1.5),flow rate 0.8 ml·min~ -1 . Diazepam(DZP)was the internal standard.Ultraviolet detection wavelength was 254 nm,AuFs=0.02. Results The linear was fine.The intra-day and the inter-day precision were both less than 8% respectively. Conclusion The method is rapid,sensitive and practical.
