摘 要:目的探讨正常与受照射的新生儿脐带血中ACl33+细胞的分离与纯化的相关问题。方法采用免疫磁珠活性分选系统(MidiMACS)分离、纯化脐带血,通过流式细胞仪测定ACl33+细胞的纯度。结果在正常与受照射的脐带血单个核细胞(MNC)中,ACl33+细胞所占的比例分别为(0.93±0.20)%和(0.33±0.20)%,两者相比较,差异非常显著(P<0.01);正常与受照组一次过柱获取的ACl33+细胞的纯度分别为(70.3±4.9)%和(68.3±4.6)%,二次过柱获取的ACl33+细胞的纯度分别为(86.3±5.5)%和(84.5±6.8)%,正常与受照组相比较,无论是一次过柱还是二次过柱,差异均无显著性意义(P>0.05)。结论免疫磁珠活性分选技术是一套有效的分离、纯化系统,可以获得足够数量的造血干/祖细胞;正常与受照射的脐带血MNC中ACl33+细胞所占的比例差异非常显著。Objective To investigate the problems in relation to the isolation and purification of normal and 6 MV-X ray irradiated umbilical cord blood AC133^+ cells.Methods The umbilical cord blood AC133^+ cells were isolated and purified by MACS purification system and the purity of AC133^+ selected cells was analyzed by flow cytometry.Results The irradiated umbilical cord blood mononuclear cells contained AC133^+ cells at a proportion of 0.33%±0.2 SEM and the normal umbilical cord blood contained AC133^+ cells at a proportion of 0.93%±0.2 SEM,and there is significant difference between them (P<0.01).For the two groups, if cells were eluted only once through the column, The normal and irradiated umbilical cord blood contained 70.3%±4.9 SEM and 68.3%±4.6 SEM of AC133^+ cells respectively. However, the percentage of AC133^+ cells increased to 86.3%±5.5 SEM and 84.5%±6.8 SEM when cells were eluted twice through the column. There was no signifi cant difference between the two groups. Conclusions MACS system is a effective isolation and purification system and we can acquire enough hematopoietics stem/progenitor cells by this system. There are significant differences between the percent of normal and irradiated umbilical cord blood mononuclear AC133^+ cells.
