风疹病毒母婴传播的定量PCR分析

摘　　要：目的研究风疹病毒(rubellavirus,RV)母婴传播的量化关系。方法应用荧光定量PCR方法检测23例IgM抗体阳性的孕妇及其婴儿血清中RVRNA含量。结果孕妇RVRNA含量与婴儿RVRNA含量呈正相关,当孕妇血清中病毒含量在7.93×102±2.03×102拷贝/μl时,婴儿血清中没有检测到RV;当孕妇血清中RVRNA含量在3.89×103±1.10×103拷贝/μl时,婴儿血清中可以检测到RV。并且随着孕妇血清中RVRNA含量的升高,婴儿血清RVRNA含量相应增高。结论孕妇与婴儿血清中RVRNA含量呈正相关。孕妇血清中RVRNA含量在3.89×103±1.10×103拷贝/μl时可能是导致婴儿感染的阈值,这对指导临床有一定的理论意义和实用价值。Objective To study the quantitative relationship of the Rubella Virus(RV) loads between the RV-infected gravidas and their infants during pregnant transmission. Method The levels of RV RNA in the serum samples of 23 couples of infants and gravidas, who were detected as RV-IgM antibody positive, were exmained by the fluorogenic quantitative real-time PCR. Result The RV RNA loads in the serum of the gravidas was positive correlated with that of the infants. When RV RNA loads in the serum of the gravida were about 7.93×10~2±2.03×10~2 copies/μl, RV RNA was not able to be detected in the serum of the infants. When RV RNA loads of the gravida were about 3.89×10~3 ± 1.10×10~3 copies/μl, RV was not able to be deteced in the serum of the infants. And the levels of RV RNA in the serum of the infants incresased with that of the gravidas. Conclusion The RV RNA loads in the serun of the gravidas and that of the infants have a positive correlation. The level of 3.89×10~3±1.10×10~3copies/μl of the RV RNA in the serum may be used as the threshold to judge if a gravida can transmit RV to the infant during pregnancy. This has some clinical values on theory and in practice.
