摘  要:目的观察CD137分子对衰老小鼠脾脏T细胞活化的影响,探讨该分子对衰老T细胞功能的调节作用。方法通过注射D半乳糖建立亚急性衰老小鼠模型。分离青龄组、自然衰老组、对照组、衰老模型组小鼠的脾脏T细胞,分别用ConA或ConA+CD137单抗活化24h,采用流式细胞术测定T细胞的凋亡率,ELISA测定细胞培养上清IL2浓度。结果自然衰老组及衰老模型组小鼠的脾脏T细胞静息培养及活化培养后的凋亡率均显著高于青龄组及对照组;ConA+CD137单抗活化组的衰老T细胞的凋亡率低于ConA活化组,其培养上清IL2浓度高于ConA活化组。结论CD137作为T细胞活化的共刺激分子能促进衰老T细胞活化后的存活及IL2的分泌,这有助于提高衰老T细胞的功能。Objective To observe the effect of CD137 on the activation of spleen T lymphocytes of senile mice and explore its regulative roles. Methods Subacute senile mice model was established by injection of D-galactose. The spleen T lymphocytes of young group, natural aging group, control group and model group were isolated and stimulated in vitro with ConA or ConA+CD137mAb for 24 hours. The apoptotic rates of T lymphocytes were detected by flow cytometry. Then IL-2 concentrations in supernatant of cells cultural were determined by ELISA. Results T lymphocytes apoptosis rates of aged and D-galactose model group were higher than those of young and control group. The apoptotic rates of aged T lymphocytes stimulated with ConA+CD137mAb were lower than those with ConA, and IL-2 concentrations of aged T lymphocytes stimulated with ConA+CD137mAb in cell culture supernatants were higher than those with ConA. Conclusions CD137 as a costimulatory molecular could promote the survival of aged T lymphocyte and increase IL-2 secretion, which help improve the function of aged T cell.
