摘 要:目的 研究氟对大鼠肝细胞DNA的损伤作用以及硒和维生素C对氟致大鼠肝细胞DNA损伤的影响。方法 氟 (F)组、氟 +硒 (F +Se)组、氟 +维生素C(F +VC)组分别用NaF 15 0mg/L、NaF 15 0mg/L +Na2 SeO35mg/L、NaF 15 0mg/L +VC 10 0 0mg/L喂养雄性SD大鼠 4周 ,应用单细胞凝胶电泳技术 (SCGE)检测肝细胞DNA的损伤率。结果 F组肝细胞DNA损伤率 (44 .80 % )明显高于对照组 (9.40 % ) ;而F +Se组(12 .60 % )和F +VC组 (14.60 % )的DNA损伤率明显低于F组 ,与对照组差异无显著意义。结论 过量氟摄入可导致大鼠肝细胞DNA损伤 ,而适量的硒和维生素C对氟致DNA损伤有一定的拮抗作用。Objective To study the effects of fluoride on hepatocyte DNA damage and the effects of selenium and VC on hepatocyte DNA damage caused by fluoride in rats.Methods Four groups of male SD rats were provided with deionized drinking water containing 0, F(150 mg/L NaF), F+Se(150 mg/L NaF + 5 mg/L Na 2SeO 3), F+VC(150 mg/L NaF +1000 mg/L VC) respectiverly for 4 weeks. Single cell gel electrophoresis assay was used to test the DNA damage rate in rat liver cells.Results The DNA damage rate(44.80%) of fluoride exposed group was significantly higher than that of the control group(9.40%), the F+Se group(12.60%) as well as the F+VC group(14.60%). The DNA damage rate of the latter two groups were not significantly defferent from the control group.Conclusions The data indicated that high dose fluoride could induce hepatocyte DNA damage in rats, and the properly selenium and VC could antagonize the DNA damage caused by fluoride.
