EFFECT OF DIFFERENT DOSES OF METHIMAZOLE ON HUMAN THYROID PEROXIDASE IN VIVO

摘  要:应用愈创木酚法对67例术前服0~40mg/d他巴唑(MMI)的Graves病甲状腺过氧化物酶(TPO)活性进行测定。MMI为0~15mg/d时,随剂量增加TPO活性渐降低;当到15mg/d时,TPO活性基本降至最低,即使再增加剂量,TPO活性未再继续降低。MMI顿服与分服相同剂量时TPO活性相同,对TPO抑制作用至少可持续24小时。因此MMII5mg/d是治疗甲亢的合适剂量,每日1次顿服的疗法是合理而有效的。The activities of thyroid peroxidase (TPO)were measured by means of guaiacol assay in 67 patients with Graves disease who had been treated witb 0~40 mg/day methimazole(MMI)preoperatively.With dose of MMI from 0 to 15 mg/day ,TPO activity de-creased with increasing dose,and reached a minimum at the dose of 15 mg/day, but without further decrease even with larger dose than 15 mg/day.There was no difference of TPO activity between patients taking the drug in divided doses and those taking one sin-gle dose of the same amount ,nor between patients taking the final dose 2 hours before thyroid excision and those taking it 26 hours before excision. Our results indicate that MMI 15 mg/day has the greatest inhibitory effect on TOP in vivo ,and inhibitory effectof MMI on TPO may last for 24 hours. The single daily dose of MMI is a reasonable and effective treatment for hyperthyroidism.
