摘  要:本实验用胶原酶消化法分离新生大鼠胰岛,培养和Ficoll密度梯度离心纯化胰岛。用海藻酸-聚赖氨酸-海藻酸生物膜包裹胰岛并进行体外胰岛素分泌功能测定和体内异种移植实验。10只链脲霉素诱导的糖尿病小鼠腹腔内移植微囊化胰岛3天后,小鼠血糖降至正常,体重逐步增加,糖尿病得到纠正。维持最长者达130天,平均85.2天。明显优于单纯胰岛移植组,其功能维持不足9天。肾脏病理组织染色和透射电镜显示早期微囊化胰岛移植确能防止糖尿病并发症的发生。Islets were isolated from SD neonatal(1-5days)rat Pancreas by a standard coll-agenase (type I sigma)diGestion techniqne and were purified by culture and ficoll Discontinuous density gradient centrifugation. The neonaTal rat islets were encapsulatid in a protective biocompatiable alginate-polysine alginate membrane. The islets secrete function nas measured in vitro and the xenotrans plantation of the islets was performed in ten diabetic mice (C57BL6, male, body weight about 18- 20 grams) induced by streptozotocin (1% ip. 220 mg/kg).The intraperitoneal transplanted encap sulated islets reversed the diabetic state of the recipients within 3 days.The blood glucose kept normal and the body weight increased gradually. For up to 130 days, with a mean xenograft survival of 85 days. This was sigifcantly different from nonencapsulated isletswhich only functioned within 9 days. No curative offect was oberved in the group transplanted with no-islet-microcapsccle. Renal pathology and electric microscopy showed that encapsulated islets xenograft can prevent diabetic complications in the early stage of the disease.
