摘  要:目的了解不同组织端粒酶活性随年龄变化的规律,为治疗性克隆核供体细胞的选择提供依据。方法采用PCR-ELISA法检测不同年龄SD大鼠皮肤、肌肉、骨髓、脑和睾丸组织的端粒酶活性。结果(1)随着年龄的增长,各组织端粒酶活性均呈下降趋势;(2)新生皮肤、肌肉和脑组织表达端粒酶活性,一月后则不再表达端粒酶活性;(3)一月至二年骨髓和睾丸组织均保持较高端粒酶活性。结论端粒酶活性表达程度与不同组织有关,骨髓和睾丸组织能维持较高端粒酶活性;骨髓组织中持续表达端粒酶活性的细胞可能是一种较好的核供体细胞。Objective To explore the telomerase activity in different tissues of rats at different ages and provide references for the choice of nucleus donor cells used in therapeutic cloning. Methods PCR-ELISA assay was used to detect the telomerase activities in skin,muscle,bone marrow,brain and testicle tissues in rats at different ages. Results (1) The telomerase activities in different tissues decreased with the growing ages. (2) The telomerase activities were expressed in the skin,muscle and brain tissues in the neonatal rats and not found in other age groups. (3) The telomerase activities in bone marrow and testicle were high in ages ranged from 1 month to 2 years. Conclusion (1) The expression of telomerase activity varied with the different tissues. The telomerase activity is higher in bone marrow and testicle tissues; however,it is not found in skin,muscle and brain tissues soon after birth. (2) The telomerase activity is negatively related with age. (3) The cells in bone marrow which can sustain higher expression of telomerase activity may be a kind of promising nucleus donor cells for therapeutic cloning.
