摘 要:目的:给予粉尘螨抗原致敏小鼠分别灌服粉尘螨抗原PLGA微球(直径7~10 μm)和粉尘螨(浸液)抗原脱敏或减敏治疗疗效的观察,以选择一种对尘螨性哮喘脱敏治疗的药物.方法:粉尘螨抗原致敏C57-BL/6小鼠,灌服粉尘螨抗原PLGA微球以及等剂量的粉尘螨(浸液)抗原减敏后,检测小鼠BALF和血清中的弹性蛋白酶、粉尘螨特异性IgG、粉尘螨特异性IgE,以及血清中的IL-4、IFN-γ,并对支气管肺组织进行病理学检查.结果:粉尘螨抗原致敏小鼠血清中粉尘螨特异性IgE、IL-4水平明显升高,粉尘螨特异性IgG水平和IFN-γ含量明显降低,BALF中弹性蛋白酶活性、粉尘螨特异性IgE水平升高,与正常对照组相比具有显著性差异;致敏小鼠气道中产生大量炎性渗出液.灌服粉尘螨抗原PLGA微球减敏后,血清中粉尘螨特异性IgE、IL-4水平下降,粉尘螨特异性IgG水平、IFN-γ含量明显升高,BALF中弹性蛋白酶活性、粉尘螨特异性IgE水平明显降低,与粉尘螨抗原致敏模型组相比具有显著性差异(P<0.05);气道中渗出液明显减少,粉尘螨特异性IgG水平无明显差异;而灌服等剂量的粉尘螨(浸液)抗原后对致敏小鼠无明显疗效.结论:直径为7~10μm、10 μg/10 g体重的粉尘螨抗原PLGA微球对致敏小鼠具有减敏作用,是一种新型的治疗粉尘螨过敏性哮喘的口服脱敏缓释制剂.Objective:To investigate the effect single administration of Dermatophagoides farinae (Der.f)-PLGA microspheres on sensitized mice. The Poly lactic-co-glycolic acid (PLGA 75/25)-encapsulated Der.f antigen and the Isodose Der.f antigen extracted were fed singly to animals 2 weeks after immunization.Methods:C57-BL/6 mice sensitized by Der.f antigen were fed with PLGA-encapsulated Der.f antigen (diameter was 7~10μm approximately) and the isodose Der.f antigen extracted, the Elastase-like activity, anti-Der.f specific IgG,anti- Der.f specific IgE, IL-4 and IFN-γ in Bronchoalveolar Lavage Fluid(BALF)or serum were assayed, and broncho-pulmonary tissues were observed histopathologically. Results: After C57-BL/6 mice were sensitized by Der.f antigen extract,anti-Der.f specific IgE and IL-4 in serum went up, the Elastase-like activity and anti- Der.f specific IgE in BALF increased, anti-Der.f specific IgG and IFN in serum decreased, and there was significant difference in the normal control group.Inflammatory exudates in airway increased obviously. After hyposensitized by the Der.f-PLGA microspheres, anti-Der.f specific IgG and IFN-γin serum increased significantly, the level of the Elastase-like activity,anti- Der.f specific IgE in BALf declined and there was significant difference.And inflammatory exudates in airway decreased obviously.But no effect for the mice fed with isodose Der.f antigen extracted.Conclusion: PLGA microspheres may serve as a powerful vehicle to improve the tolerance effect of the PLGA Der.f microspheres, and single administration of PLGA Der.f microspheres may hold promise as a new treatment strategy in desensitizing Der.f sensitized disease.
