Determination of Trace Antimony in Cells by Inductively Coupled Plasma Mass Spectrometry for drug resistance study of Protozoan Parasite

摘  要:为了研究利什曼原虫(Leishmania)对于含五价锑药物的抗药性机理而建立了一种测定细胞内痕量金属锑的方法。本文尝试使用浓硝酸冷消化法处理细胞样品,简化了实验步骤,降低了被测物质损失和污染的可能性。电感耦合等离子体-质谱(ICP-MS)作为检测手段令本方法测定金属锑的最低检出浓度达到0.036μg?l-1,线性范围达到1.0-80ng?ml-1,加标回收率为98.0-110.0%,日间精密度≤1.63%。本方法较为简单,结果准确可靠,已经应用于寄生虫的金属抗药性研究,所获结果完全符合预期的实验模型。An analytical method for determination of antimony (Sb) in parasite cells using inductively coupled plasma mass spectrometry (ICP-MS) was established in order to study the mechanism of metal resistance in the protozoan parasite Leishmania. The cell samples were treated by cold digestion, which simplified the experimental procedures and reduced the possibility of sample loss and that of contamination. The detection limit and the between-day RSDs were 0.036 μg·1-1 and 1.6%, respectively. The average recoveries at different concentrations were 98.0-110.0%. This method was simple, precise,reliable,and has been successfully applied to the study of the mechanism of metal resistance in Leishmania. The obtained results were fitted well with the predicted model.
