猪胰岛移植治疗大鼠药物性糖尿病的研究

摘　　要：本研究观察了猪胰岛移植于糟尿病大鼠肝内逆转其高血糖状态的效果，结果发现１５只移植鼠均存活，无一出现急性门静脉高压、肝梗死等并发症，其中１２只受鼠高血糖状态得到暂时缓解，最低血糖值从３６０．９０ｍｇ／ｄｌ±５６．４ｍｇ／ｄｌ降至１６５．８０ｍｇ／ｄｌ±７８．３８ｍｇ／ｄｌ，维持血糖正常平均达５．６天±２．４天，而糖尿病对照组病鼠在观察期间无一只自行缓解。认为异种胰岛移植于肝内是安全可行的，移植的胰岛能存活并可发挥功能。Abstract 27 Wistar rats with Strptozotozin-induceddiabetes were divded into two groups.Group1:untreated diabetic control rat(n=12).Group 2:transplanted rats(n=15).The pigislets were cultured at 24 ℃ for 7 days,1200-1500 islets were transplanted intraportally in-to each diabetic rat; immunosuppression thera-py wasn't used.The result showed that plasmaglucose remained in hight lever in any rat ingroup 1,the symptom didn't resolve.in group2,acute portal hypertension、liverinfarct、DICwere not occurred after islet xenotrans-plantation;the plasma glucose in 12 rats of 15returned to normoglycemia and the normo-glycemia was meanly remained for 5.6 days,the longst Was 9 days. Our result shows thatthe intraportal transplantation of xenogeneticislets was Well tolerated in the rat;the trans-planted islets can survive and reverse hyper-glycemic condition in the diabetic rats.
