乳糖发酵奈氏菌和脑膜炎奈氏菌诱生的流脑杀菌抗体抗原特异性研究

摘　　要：利用杀菌力试验测定不同来源血清对脑膜炎奈氏菌（Ｎｍ）的杀菌抗体水平。结果表明Ａ群流脑病人恢复期血清、带菌者及乳糖发酵奈氏菌（Ｎｌ）带菌者血清、兔抗Ｎｍ及Ｎｌ血清中均具有对Ｎｍ的杀菌抗体。通过Ｗｅｓｔｅｒｎｂｌｏｔｔｉｎｇ分析发现上述人血清ＩｇＧ、ＩｇＭ和ＩｇＡ均与这两种奈氏菌的５２ｋＤ，１、２或３类外膜蛋白发生特异反应。用纯化的１和３类外膜蛋白及脂寡糖免疫小鼠制备免疫血清，测定抗体水平表明，Ｎｌ、Ｎｍ的１和３类外膜蛋白及脂寡糖抗原对小鼠具有一定的免疫作用；其中１类外膜蛋白主要诱生ＩｇＧ，３类外膜蛋白诱生ＩｇＧ和ＩｇＭ，脂寡糖则主要诱生ＩｇＭ。通过杀菌抑制试验证实１类、３类外膜蛋白及脂寡糖均可抑制ＩｇＧ的杀菌作用；抑制效价ＬＯＳ＞１类ＯＭＰ＞３类ＯＭＰ。由此可见，Ｎｌ和Ｎｍ的脂寡糖及１类ＯＭＰ是幼儿咽部携带Ｎｌ或Ｎｍ诱生的流脑杀菌抗体主要特异性抗原。这一研究结果为提高流脑Ａ群多糖菌苗对婴幼儿的预防效果提供了线索。o identify the antigenic specificity of meningococcal bactericidal antibodies the bactericidal antibodies in seracollected from different sources have been detected.The results indicated that the bactericidal antibodies Vs.Nmgroup A were found in the sera of carriers of N. lactamica ,rabbit immunized with Nl 1105,the convalescent sera ofcases of epidemic menin8itis and the sera of carriers of N.mening itid is serogroup A as well as the sera of rabbit im-munized with N.mening itid is serogroup A. IgG,IgM and IgA purified from the above sera specifically reactedwithclass 1,2/3 OMP,Moreover,class 1,3 OMPs and LOS purified from Nl 1105 were found to induce good im-mune response to N.meningitid is serogroup A in the Kunming strain of white mice:class 1 OMP could induce IgG,class 3 OMP induced both IgG and IgM ,whereas LOS mainly stimulated formation of IgM.In addition ,the resultsdemonstrated that class 1,3 OMPs and LOS purified from Nl 1105 could inhibit the bactericidal activity of IgG. Thecomparative inhibitory activity was as following : LOS>class 1 OMP>class 3 OMP.From the above findings,Weconsider that LOS and class 1 OMP of N.lactamica seem to be major specific antigens inducing the bactericidal an-tibodies against meningococci.These results provide important information for raising prophylactic effect ofserogroup A meningococcal vaccine to infants.
