益气解毒颗粒对HNE3细胞及其裸鼠移植瘤的抑制作用

摘　　要：目的观察益气解毒颗粒对鼻咽癌细胞HNE3及其裸鼠移植瘤的抑制作用。方法益气解毒颗粒作用于HNE3细胞、宫颈癌细胞及其裸鼠移植瘤,分别进行体外抑瘤试验(包括IC50测定)和体内抑瘤试验。先行体外实验测定益气解毒颗粒对癌细胞半抑制浓度及其杀伤效力,再行体内实验,观察益气解毒颗粒对荷瘤裸鼠HNE3移植瘤的抑瘤效应,并以人宫颈癌细胞HeLa移植瘤和荷瘤模型动物作对照。结果益气解毒颗粒对HNE3细胞的半抑制浓度为0.037mg/mL,并显示了明显的体外杀伤效应和细胞凋亡诱导效应。在体内抑瘤实验中,益气解毒颗粒对HNE3移植瘤的抑瘤率为74.7%,伴有细胞增殖速度、成瘤能力的明显抑制,同时提高细胞分化能力,降低肿瘤的侵袭力。结论益气解毒颗粒对鼻咽癌细胞HNE3具有明显抑制作用。Objective To investigate the inhibitory effects of Qi-boosting Toxin-resolving granule (QBTRG) on nasopharyngeal carcinoma (NPC) cells HNE3.Methods Experimental studies were carried out in cell lines of HNE3 and HeLa, the cell lines of NPC and cervical carcinoma respectively, and in nude mice with transplanted tumors of HeLa cells or HNE3 cells. The semi-inhibitory concentration (IC_(50)) of QBTRG was determined in vitro in the experiments with these two cell lines at first. Then, the inhibitory effects of this medicine on these cell lines were assayed in the concentration around IC_(50). On the other hand, tumor-growing curve and tumor-inhibiting rate were determined in vivo among animals with transplanted tumors of NPC or cervical carcinoma cells, following the treatment of QBTRG. Results IC_(50) of QBTRG was 0.037mg/mL and it held significant inhibiting effects on these tumor cells in vitro, not only bringing about apoptosis-inducing effect but also giving rise direct killing effect on them. As seen in the experiments in vivo, QBTRG could bring about significant inhibiting effects on transplanted tumors of HNE3 cells or HeLa cells as well, slow down the proliferating rate of these cells, inhibit the ability of transplanting tumor-forming, promote differentiation of them, reduce invading potentiality of the cells and improve the general condition of the tested animals. The tumor-inhibiting rate was 74.7% for HNE3 cell transplanted tumors. Conclusion QBTRG holds very significant inhibitory effects on NPC cells. 〔
