摘  要:目的探讨β—榄香烯对微管蛋白体外聚合作用的影响,阐明其抗肿瘤的作用机制。方法采用MTT法测定β—榄香烯对体外培养的A549、WI38细胞增殖、生长的抑制作用;用浊度法测定β—榄香烯对提取的牛脑微管蛋白体外聚合的抑制作用。结果β—榄香烯能明显抑制A549、WI38细胞增殖、生长,其抑制作用呈浓度与时间依赖性;对A549、WI38细胞增殖的半数抑制率(IC50)分别为2.73μmol/L、3.53μmol/L。β—榄香烯明显抑制体外微管蛋白的聚合,并与药物浓度呈一定的线性关系,对微管蛋白体外聚合的半数抑制率(IC50)为5.12μmol/L。结论β—榄香烯对A549、WI38细胞增殖、生长有明显的抑制作用,β—榄香烯抑制微管蛋白的聚合是其抗肿瘤作用的重要机制之一,并与微管蛋白稳定剂紫杉醇的作用机理不同。Objective To study the mechanism of β-Elemene in antitumor by analyzing the polymerization of tubulin in vitro.Methods The inhibition of β-Elemene on proliferation and growth of cultivated cells A549 and WI38 were determined with MTT and the inhibition of β-Elemene on polymerization of tubulin isolated from bovine was determined with turbidimetry. Results Both proliferation and growth of cultivated cells A549 and WI38 and the poly werization of fubulin isolated from bovine were inhibited evidently.The IC50 for the former were 2.73μm and 3.53μm and for the latter 5.12μm.Conclusions β-Elemene can inhibite the proliferization and growth of cultivated cells A549 and WI38; the inhibitory effect on tubulin polyrnerization may be one of the onechanisms of antitumor.
