摘  要:目的建立高效液相色谱法测定决明子中大黄素和大黄酚含量的方法。方法采用HP1100高效液相色谱仪(包括G1314A可变波长紫外检测器,HP化学工作站,G1322A真空脱气机,HP1100四元泵),HypersilODS色谱柱(4.6mm×250mm,5μm)。流动相:甲醇-0.1%磷酸溶液(85∶15);紫外检测波长:254nm;流速:1.0mL/min;进样20μL。结果大黄素在0.02~0.4μg范围内呈现良好的线性关系,大黄酚在0.04~0.8μg范围内呈现良好的线性关系,大黄素回收率为98.46%(n=5),RSD=1.34%。结论该法简便、准确、具有专属性,可用于测定决明子中大黄素和大黄酚的含量。Objective To establish a HPLC method for analysis of Emodin and Chrysophanol in Semen Cassiae. Method The HPLC system consists of G1314A variable wavelength uv-visible detector, Hypersil ODS (4.6 mm×250 mm, 5 μm), HP chemstation, HP1100 Series Quaternary Pump and Vacuum degasser (G1322A). The mobile phase was methanol-0.1% Phosphoric acid (85∶15, v/v). The flowrate was 1.0 mL/min, the injection volume was 20 μL, and the detector wavelength was 254 nm. Result The method was linear of Chrysophanol at the range of 0.04~0.8 μg. The method was linear of Emodin at the range of 0.02~0.4 μg. The recovery was 98.46% (n =5), RSD was 1.34%. Conclusion The method was simple and accurate, it can be used for the determination of Emodin and chrysophanol in Semen Cassiae.
