特发性血小板减少性紫癜患者血浆中血小板颗粒膜蛋白－140自身抗体的研究

摘　　要：应用单克隆抗体ＳＺ－５１（ＥＬＩＳＡ法）对９２例特发性血小板减少性紫癜（ＩＴＰ）患者的血浆检测了血小板颗粒膜蛋白（ＧＭＰ）－１４０自身抗体，结果显示１７例（１８，５％）血浆中存在ＧＭＰ－１４０自身抗体。用亲和层析纯化的ＧＭＰ－１４０作ＳＤＳ－ＰＡＧＥ后免疫印迹，分别与８例ＩＴＰ血浆反应，结果有２例在分子量１４万处显示阳性区带。５９例检测结果示ＧＭＰ－１４０自身抗体常与其它血小板糖蛋白自身抗体同时存在（９１．７％）。lasma from 92 patients with chronic idiopathicthrombocytopenic purpura (ITP) was tested for autoan-tibodies directed against granule membrane protein 140(GMP-140) with antigen--specific immunoassay. Re-sults showed that autoantibodies to GMP-140 existed in17 patients with ITP (18. 5%). After SDS-PAGE ofpurified GMP-140 immunoblotting showed that 2 out of8 sera from patients presented a staining band with M.W. of 140 Kd. 59 plasma samples were subjected to de-tection of autoantibodies to GMP-140, GPⅡb/Ⅲa andGPⅠb/Ⅸ, simultaneously. It was shown that most ofpatients presented anti-GMP-140 activities coexistedwith anti-GPⅡb/Ⅲa and/or anti-GPⅠb/Ⅸ ( 91. 7%).Our study provided direct evidence for autoantibodies toGMP-140 in some of patients with ITP. It has certainsignificance for further study on the pathogenesis ofITP.
