摘 要:目的探讨内源性一氧化碳(CO)和一氧化氮(NO)在糖尿病(DM)大鼠高血压发病中的相互作用。方法监测DM大鼠动脉血压,测定下肢动脉环对苯肾上腺素(PE)累积收缩反应和对乙酰胆碱(ACh)累计舒张反应,检测血中CO和NO含量。结果DM大鼠出现低体重、高血压和高血糖,下肢动脉累积收缩和舒张反应明显降低,血中CO含量明显降低,诱导型一氧化氮合酶(iNOS)活性和NO水平则明显上升;应用正铁血红素(He)可部分逆转DM高血压,明显改善下肢动脉的收缩反应和舒张反应,提高血中CO水平,同时抑制iNOS活性和NO合成。结论DM大鼠早期内源性CO合成减少和代偿性NO合成增加共同参与诱导下肢血管反应障碍,是引起DM高血压的重要机制之一。Objective To explore the effects of both endogenous carbon monoxide(CO) and nitric oxide (NO) on the pathogenesis of hypertension of diabetic rats.Methods The changes of arterial blood pressure were observed.Using isolated vascular ring tension detecting technique,cummulative relaxation responses to acetylcholine(ACh) and contraction responses to phenylephrine (PE) of lower limb arteries were measured.The content of both CO and NO in blood was detected.Results Diabetic animals expressed lower body weight,higher blood glucose and increased blood pressure.Both of the cummulative dose relaxation responses to ACh and contraction dose responses to PE were decreased in diabetic rats.Pretreatment with hemin in diabetic rats could partly reverse the vascular hyporeactivity,the level of CO was declined.Pretreatement with hemin in diabetic rats could decrease the level of NO and activity of iNOS.Conclusion Both endogenous CO and NO are involved in regulating vascular tone of the lower limb arteries in early stage of diabetic rats,and they may be compensated each other for regulating the pathogenesis of hypertension,which may be one of the important mechanisms of diabetes company hypertension.
