摘 要:将处理后的人羊膜夹在改进的Boyden小室中间,于羊膜绒毛膜面培养人血管内皮细胞,建立了稳定的血管内膜培养模型。以巯基氧化剂联胺作用于内皮细胞,引发细胞过氧化脂质蓄积与脂质过氧化损伤。此内皮细胞损伤能增加单核细胞对内皮细胞的粘附和内皮下穿入;单核细胞主要粘附于内皮细胞间隙与受损伤细胞表面;在内皮细胞剥脱区域穿入内皮下的单核细胞较非剥脱区域为多。结果提示,内皮细胞脂质过氧化损伤通过增加单核细胞的粘附与内皮下穿入而在动脉粥样硬化发生中起重要作用。he influnces of lipid peroxidative injury toendothelial cells (EC) on adherence to EC and migra-tion into subendothelial space of human monocyteswere studied by using a modified vascular endothelialmodel in vitro. Lipid peroxidative injury to culturedhuman umbilical vein EC was initiated by treating theEC with diamide. The results showed that injury to ECcould enhance adherence to EC and migrition intosubendothelial space of monocytes. Monocytes mainlyadhered to the intercellular space and surface of in-jured EC. In the area of EC denudation, the numberof monocytes which migrate into the subendothelialspace was more than that in the area withoutdenudation. These results suggested that enhanced ad-herence to EC and rnigntion into subendothelial spaceof monocytes caused by lipid peroxidative injury toEC may play an important role in atherogenesis.
