摘 要:用细胞液流室和显微录像技术观察重症烧伤大鼠白细胞在不同切应力下与鼠肺血管内皮细胞粘附特性的变化。发现重症烧伤后3小时,白细胞粘附于内皮细胞的数目明显增多,粘附曲线上移,而且,白细胞粘附率与动物存活时间明显相关。给予虎杖4号可明显降低白细胞粘附率,粘附特性曲线恢复正常,动物存活时间显著延长。本实验结果证明,重症烧伤后大鼠的白细胞粘附特性有明显增强。wenty-four rats were divided into 3 groups:normal controls (8), simple bumed (8), bumed plusPolygonum cuspidatum (p.c.)-treated (8). Three hoursafter burn, 3ml of whole blood was drawn to isolatepolymorphonuclear neutrophilic leucocytes (PMNs).Confluent monolayer of rat microvessel endothelialcells was grown in a parallel plate of flow chamber.PMNs were driven through the flow chamber underdifferent wall shear stresses. Leukocyte-endotheliaadhesion was observed with an Olympus inverted mi-croscope and recorded with a colour microscopic TVset. the adhesive characteristic curve of leukocyte wassignificantly different between the nomal and burnedgroups. The number of PMNs adhesive toendothelium increased obviously 3 hours after burn.Under 0. 196 Pa of wall shear stress, the adhesion rateof PMNs was 81.2+4.1% in the burned group and58.8+4.6% in the normal group. The PNMs adhesionrate was decreased to near normal in p.c. -treatedgroup. The average survival time in the burned groupwas 4.1+1.2 hours, much lower than that in the p.c.treated group (33.2+1 5.8 hours), However, in the sim-ple burned group, one rat survived for 24 hours and itsPMNs adhesive curve was similar to that of the nor-mal or p. c.-treated groups. The findings indicate thatPMNs adhesion features increase apparently after se-vere burn, which is closely ralated to the survival rate.
