摘  要:目的:观察动脉粥样硬化兔血管内皮功能及乐卡地平的干预效果。方法:实验于2003-03/10在中国医科大学完成。选择日本大耳白兔32只,随机分为4组,正常对照组,高脂对照组,低剂量乐卡地平组,高剂量乐卡地平组。各组按照不同的饲料喂养,正常对照组喂饲普通颗粒饲料,高脂对照组喂饲高脂饲料(100g普通饲料中加入1g胆固醇粉+7.5g蛋黄粉+2g猪油),低剂量乐卡地平组喂饲高脂饲料+乐卡地平1mg/(kg·d),高剂量乐卡地平组喂饲高脂饲料+乐卡地平10mg/(kg·d)共喂养12周。在实验前及喂养4,8,12周后采集耳静脉血,测定家兔血清脂质、一氧化氮及血浆内皮素含量。实验12周后腹腔麻醉动物,记录血压,随后处死动物,取降主动脉进行苏木精-伊红染色观察病理形态学变化;免疫组化染色观察血管细胞黏附分子1的表达分布。结果:实验过程中有2只动物死亡,30只进入结果分析。①病理形态学改变:正常对照组主动脉未见异常改变,高脂对照组动物动脉管壁呈弥漫性隆起,内膜明显增厚,可见斑块环管腔大部甚至全周,斑块内可见大量泡沫细胞及多层排列,内皮细胞缺失或不连续,平滑肌细胞排列紊乱。低剂量乐卡地平组内膜增厚程度及病变范围较高脂对照组略有减轻,高剂量乐卡地平组仅见局限性病灶隆起,内膜下可见3~4层泡沫细胞,其余动脉内膜光滑完整。②血脂水平:实验12周后各组高密度脂蛋白胆固醇、三酰甘油水平基本一致。③内皮素和血清一氧化氮水平:实验后12周高剂量乐卡地平组血浆内皮素低于高脂对照组,血清一氧化氮高于高脂对照组;低剂量乐卡地平组与高脂对照组比较无显著差异。④血管细胞黏附分子1表达:低剂量乐卡地平组、高剂量乐卡地平组血管细胞黏附分子1阳性细胞面积明显低于高脂对照组犤(19.93±6.69)%、(12.80±4.09)%,(30.12±9.95)%,P<0.05犦。⑤实验12周后进行有�AIM:To observe the vascular endothelial functions of rabbits with atherosclerosis,and investigate the interventional effect of lercanidipine.METHODS:The experiment was carried out in China Medical University from March to October 2003.Thirty-two Japanese white big-ear rabbits were randomly divided into four groups:normal control group(fed with common granule feed),high fat control group(fed with high fat diet,in which 1 g cholesterol powder,7.5 g yolk powder and 2 g lard were added to 100 g common granule feed),low and high dose lercanidipine treated groups(treated with high fat diet and lercanidipine of 1 and 10 mg/kg per day respectively),all the rabbits were fed for 12 weeks.Venous blood was drawn from the ear of rabbits before experiment and 4,8 and 12 weeks after feeding respectively to detected the contents of lipid and nitric oxide in serum and endothelin in plasma.All the rabbits received intraperitoneal anesthesia 12 weeks after experiment,and the blood pressure was recorded,and then the rabbits were killed,descending aorta was obtained to observe the pathomorphological changes with haematine eosin staining, and the distribution of the expression of vascular cell adhesion molecule 1 was observed with immunohistochemical staining.RESULTS:Two rabbits died during the experiment, and 30 were involved in the analysis of results.① Pathomorphological changes:No aortic abnormal change was observed in the normal control group;In the high fat control group,there were diffusive eminences on the arterial dissepiment,the tunica intima was obviously thickened,the plaques could be observed around most even all the canal cavity,and there were plenty of foam cells,which arranged in many layers,endothelial cells were deleted or discontinuous,and the arrangement of smooth muscle cells was disorder; In the low dose lercanidipine treated group,the thickening degree of tunica intima and the damaged range were relieved as compared with those in the high fat control group;In the high dose lercanidipine treated group,only loca
