摘 要:目的:探讨2,3-丁二醇不同孵育温度改性处理戊二醛鞣制的牛心包对其防钙化性能及热皱缩温度的影响。方法:实验于2003-06/12在中南大学湘雅二医院胸心外科实验室完成。收集1~3岁新鲜黄牛心包8个,将每个牛心包均分成5份,共分成5组,分别置入4℃,20℃,30℃,40℃的纯2,3-丁二醇溶液孵育改性60d,对照组置入4℃的3g/L戊二醛Hepe缓冲液中,每组8份试片。选择刚离乳SD雄性大鼠8只,SD大鼠背部作两个纵形切口,将经过处理的同一牛心包来源的5份心包试片植入切口,第1个纵形切口两侧各1片,第2个纵形切口两侧及下部各1片,相距≥2cm,植入牛心包试片60d后,取出各组试片,用原子吸收火焰法检测组织钙含量,并观察牛心包试片热皱缩温度。结果:8只SD大鼠所获全部标本进入结果分析。①组织钙含量检测结果:20℃,30℃和40℃丁二醇组组织钙含量相似,但明显低于对照组和4℃丁二醇组犤(8.18±7.22),(21.56±26.53),(3.24±2.57),(138.36±69.23),(192.83±2.86)mg/g,(P=0.002~0.016)犦。②热皱缩温度检测结果:20℃,30℃及40℃丁二醇组明显低于对照组及4℃丁二醇组犤(83.31±0.46)℃,(82.96±0.33)℃,(82.44±0.42)℃,(83.75±0.46)℃,(83.67±0.41)℃,(P<0.05)犦;40℃丁二醇组明显低于20℃丁二醇组(P<0.001)。结论:2,3-丁二醇的不同孵育温度改性处理戊二醛鞣制的牛心包,其中4℃组防钙效果最差,20℃,30℃,40℃组均具有明显的防钙化效果,三者之间无明显差异。热皱缩温度随着孵育温度的升高而降低。但改变幅度不是很大。均高于80℃,改性后的牛心包在生理温度下不会发生组织热皱缩。AIM:To investigate the influence of bovine pericardia tanned with glutaraldehyde of different incubating temperature of 2,3-butanediol post-treatment on its anti-calcification and heat shrinking temperature.METHODS:The experiment was carried out in the laboratory of Department of Cardiothoracic Surgery,Second Xiangya Hospital,Central South University between June and December 2003.Eight fresh pericardias of 1 to 3 year old bovine were collected,each bovine pericardia was divided into 5 groups,and were placed into the 2,3 butanediol solution at 4 ℃ ,20 ℃ ,30℃ and 40 ℃ for modified incubation for 60 days,those in the control group were placed in the 3 g/L gutaradehyde Hepe buffer of 4 ℃ ,there were 8 pieces in each group.Eight weanling male SD rats were selected,two vertical incisions were made on the back of the SD rats,the 5 pieces of pericardia slice from the same bovine pericardia after treated were implanted incisions,one on the two sides of the first vertical incision respectively,one on the two sides of and below the second vertical incision respectively,the interval of each piece was ≥ 2 cm;Sixty days after the slices of bovine pericardia were implanted,those in each group were taken out;The content of calcium in the tissue was detected with flame atomic absorption spectrophotometer,and the heat shrinking temperatures were observed.RESULTS:All the samples from the 8 SD rats were involved in the analysis of results.① The calcium contents in the tissue are similar in the 20 ℃ ,30 ℃ and 40 ℃ butanediol groups,all were obviously lower than those in the control group and 4 ℃ butanediol group[(8.18± 7.22),(21.56± 26.53),(3.24± 2.57),(138.36± 69.23),(192.83± 2.86) mg/g,P=0.002 to 0.016].② The heat shrinkage temperatures in the 20 ℃ ,30 ℃ and 40 ℃ butanediol groups were obviously lower than those in the control group and 4 ℃ butanediol group[(83.31± 0.46) ℃ ,(82.96± 0.33) ℃ ,(82.44 ± 0.42) ℃ ,(83.75± 0.46) ℃ ,(83.67± 0.41) ℃ ,P< 0.05], and it was markedly lower in
