摘 要:目的:观察还少丹对老年小鼠衰老指标活性氧损害程度与DNA端粒长度的影响。方法:实验于2005-01/04在湖南中医学院分子生物学实验室完成,选择24只健康昆明种老年小鼠,随机分为对照组和药物组,各12只。药物组用还少丹水煎液(由何首乌、熟地、肉苁蓉等中药组成)灌胃10μL(含生药10g)/(g·d),对照组用等量生理盐水灌胃,每周灌胃5d,共给药70d。小鼠断头取血,离心分离出红细胞,加蒸馏水制成溶血液。溶血液中超氧化物歧化酶含量采用光化学扩增法检测,溶血液中过氧化氢酶含量以过氧化氢分解法检测,同时采血测定溶血液血红蛋白的含量。小鼠肝组织DNA的平均端粒长度应用Southern印迹杂交检测。结果:实验过程中对照组死亡3只,药物组死亡2只,进入结果分析对照组和药物组分别为9和10只。①超氧化物歧化酶活性及过氧化氢酶活性:药物组小鼠显著高于对照组小鼠犤(31.41±2.51)μkat/g,(22.95±3.69)K/g;(21.36±3.10)μkat/g,(14.12±1.96)K/g,(t=7.797,6.387,P<0.01)犦。②肝组织DNA平均端粒长度:药物组小鼠长于对照组犤(23.05±0.50)kb,(17.26±0.68)kb,(t=21.358,P<0.01)犦。结论:服用还少丹后老年小鼠超氧化物歧化酶和过氧化氢酶的活性增加,清除活性氧的能力加强,并可能通过此作用延缓了端粒的缩短,达到了延缓衰老的目的。AIM:To observe the effects of huanshao dan on the aging indexes of damaged severity of active oxygen and length of DNA telomere in mice.METHODS:The experiment was carried out in the Laboratory of Molecular Biology,Hunan College of Traditional Chinese Medicine from January to April 2004.Twenty four healthy Kunming aging mice were randomly divided into control group(n=12) and drug group(n=12).Mice in the drug group were treated with gastric perfusion of huanshao dan decoction containing 10 g crude drugs(10 μ L/g per day),which consisted of the Chinese herbs of tuber fleeceflower root,prepared rehmannia root and desertliving cistanche;Saline of the same volume was perfused intragastricly to the mice in the control group,5 days every week for 70 days.The heads of mice were cut down for blood,distilled water was added to prepare hemolytic solution after centrifugalization of erythrocytes.The activities of superoxide dismutase and catalase in hemolytic solution were detected with photochemical amplification and hydrogen peroxide resolution method;The content of hemoglobin in hemolytic solution was also determined.The average length of telomere in liver tissue DNA was detected with Southern blotting method.RESULTS:During the experiment, 3 mice in the control group and 2 mice in the drug group died,and there were 9 and 10 mice in the control group and drug group were involved in the analysis of results respectively.① The activities of superoxide dismutase and catalase were significantly higher in the drug group than in the control group[(31.41± 2.51) μ kat/g,(22.95 ± 3.69) K/g;(21.36± 3.10) μ kat/g,(14.12± 1.96)K/g,t=7.797,6.387,P< 0.01].② The average length of telomere in liver tissue DNA was longer in the drug group than in the control group[(23.05± 0.50),(17.26± 0.68) kb,t=21.358,P< 0.01].CONCLUSION:After medication of huanshao dan,the activities of superoxide dismutase and catalase are increased,and the ability in eradicating active oxygen is enhanced,and then it plays a role in delaying the shortening o
